Peptide vials and their instructions are dense with jargon: lyophilized, reconstitute, BAC water, sterile water for injection, nonpyrogenic. None of it is complicated once the words are explained, but the terms are easy to blur together, and a couple of them (which water to use) matter. This is a plain-language glossary of the reconstitution vocabulary, meant to sit alongside the fuller peptide research glossary. We’ll define each term, then show how they fit together.

Lyophilized (freeze-dried)

Lyophilized is the word you’ll see most, and it means freeze-dried. Lyophilization is a low-temperature drying process: the peptide solution is frozen solid, then placed under vacuum so the frozen water turns directly from ice to vapor (a phase change called sublimation) without ever passing back through a liquid stage. What’s left is a dry solid.

Manufacturers do this because peptides are far more stable dry than dissolved in water. Removing the water halts most of the chemical reactions that degrade a peptide, so a lyophilized powder can be shipped and stored (often refrigerated or frozen) for many months, then dissolved again just before use. The process runs in three stages: freezing, primary drying (the bulk sublimation step), and secondary drying (pulling off the last bound moisture). After a good lyophilization cycle, the peptide usually appears as a white or off-white powder.

‘Lyophilized’ means freeze-dried: the peptide was frozen and the water removed by sublimation, leaving a dry powder or ‘cake’ that stores far better than a solution.

The cake (and why it can look like nothing)

The visible solid left in the vial is often called the cake: a light, porous plug of peptide. Because research-peptide quantities are tiny (a few milligrams), the cake can be almost invisible: a thin film, a small pellet, or a powder scattered at the bottom. A vial that looks “empty” is usually normal. A collapsed, melted, or shrunken-looking cake, on the other hand, is a recognized lyophilization defect and can signal that the material was stressed. Reconstitution

Reconstitution is the act of adding a liquid back to the dry powder to dissolve it into a usable solution. That’s the entire meaning: “re-constitute” = put back together. The liquid you add (the diluent or solvent) is almost always water of some kind, and the choice of water is where the next two terms come in. Sterile Water for Injection

Sterile Water for Injection, USP is purified water that has been sterilized and contains no preservative and no antimicrobial agent. Its pH sits near neutral. Because nothing in it inhibits microbial growth, it is intended for single use — once a container is opened and exposed to air, it can be contaminated, so leftover water in an opened unit is discarded rather than reused.

Bacteriostatic Water (BAC water)

Bacteriostatic Water for Injection, USP (usually shortened to BAC water) is sterile water with one addition: 0.9% (9 mg/mL) benzyl alcohol as a bacteriostatic preservative (some presentations use 1.1%). “Bacteriostatic” means growth-inhibiting: the benzyl alcohol doesn’t necessarily kill everything, but it stops contaminating bacteria from multiplying. That property is what allows a multiple-dose vial (the label describes it as a container “from which repeated withdrawals may be made”), because the water resists spoiling between draws.

One safety note appears prominently on every BAC water label: it carries a boxed “NOT FOR USE IN NEONATES” warning, because benzyl alcohol has been linked to toxicity in newborns. That’s a real, labeled contraindication, not a marketing footnote.

The short version: the preservative is the whole difference, and it’s why BAC water is the common choice when a reconstituted vial will be drawn from many times. Nonpyrogenic and a few smaller terms

A handful of supporting words round out the vocabulary:

  • Nonpyrogenic: free of pyrogens, substances (often bacterial breakdown products) that can trigger a fever response. Injectable-grade waters are labeled nonpyrogenic.
  • Diluent / solvent: the liquid added during reconstitution (i.e., the water).
  • USP: United States Pharmacopeia, the standards body; “Water for Injection, USP” means the product meets that official monograph.
  • Concentration: how much peptide is dissolved per unit of liquid (e.g., mg/mL), set entirely by how much powder was in the vial and how much water you add.

Putting it together

A typical vial workflow reads like a sentence once you know the words: you receive a lyophilized, nonpyrogenic peptide cake; you reconstitute it by adding a diluent, either sterile water (single use) or BAC water (preserved, multi-use), which dissolves the powder to a known concentration. That’s the whole vocabulary.

Two honest caveats close this out. First, defining these terms is deterministic: the words mean what the pharmacopeial labels say they mean. Second, and more important: most research peptides are not approved for human use. Knowing what “lyophilized” or “bacteriostatic” means is educational context, not a green light.
